近日,美國南加州大學Unmesh Jadhav團隊研究了轉(zhuǎn)錄因子Cdx2在發(fā)育和穩(wěn)態(tài)過程中如何通過不同機制與基因組不同位點結(jié)合,以及這種結(jié)合如何受到DNA甲基化和染色質(zhì)修飾的調(diào)控。研究利用小鼠腸道上皮細胞作為模型系統(tǒng),通過多種實驗方法揭示了Cdx2結(jié)合的動態(tài)變化及其對基因表達的調(diào)控機制。其中,WGBS(全基因組重亞硫酸鹽測序)和ChIP-seq(染色質(zhì)免疫沉淀測序)在本研究中發(fā)揮了關鍵作用。相關研究成果以《Motif distribution and DNA methylation underlie distinct Cdx2 binding during development and homeostasis》發(fā)表于Nature子刊《Nature Communications》。
